چکیده

The cell growth on H2 and O2 as the energy source and CO2 as the sole carbon source in the autotrophic culture condition was tested for the gram-negative bacteria Paracoccus spp. The aerobic growth in the autotrophic condition was only observed in Paracoccus denitrificans NBRC13301 and P.pantotrophus NBRC102493. Both strains were sensitive to O2 in particular the growth of P.pantotrophus was completely inhibited at the concentrations above 2% O2. P.denitrificans grew until 15% O2 however the optimum O2 concentration was 5%. The growth characteristic of P.denitrificans was investigated by pH-controlled batch culture with supplying the gas mixture of H2, O2 and CO2. The specific growth rate was 0.12 h at 5% O2, 30°C and pH7.0. The growth was much slower than other hydrogen-oxidizing bacteria however P.denitrificans accumulated biodegradable plastic, polyhydroxybutyrate, PHB with the content of 57.3% w/w in the cell under nitrogen limitation. Under DO limitation, the cell concentration increased to 25 g/L without accumulating PHB if NH3 solution as the nitrogen source was fed sufficiently.

An improved medium for the anaerobic growth of Paracoccus denitrificans Pd1222

Paracoccus denitrificans is a well studied model organism with respect to its aerobic and anaerobic respiratory enzymes. However, until now, the growth medium for this organism has not been optimized for anaerobic growth. In particular, the requirements of P. denitrificans for trace elements (TEs) are not well known. In the present study we aimed to improve growth rates of P. denitrificans Pd12...

Anaerobic growth of Paracoccus denitrificans requires cobalamin: characterization of cobK and cobJ genes.

A pleiotropic mutant of Paracoccus denitrificans, which has a severe defect that affects its anaerobic growth when either nitrate, nitrite, or nitrous oxide is used as the terminal electron acceptor and which is also unable to use ethanolamine as a carbon and energy source for aerobic growth, was isolated. This phenotype of the mutant is expressed only during growth on minimal media and can be ...

Some investigations on PHA production using activated sludge as inoculum

This paper indicates that activated sludge under certain conditions can achieve levels of polyhydroxyalkanoates (PHA) accumulation which would enable the economic recovery of this precursor of biodegradable plastics. The effects of C/N ratio and temperature on polyhydroxyalkanoates production were evaluated. Acetate uptake, ammonium consumption, oxygen uptake and PHA production were studied und...

Title Production of polyhydroxybutyrate by activated sludge performing enhanced biological phosphorus removal

Recovery of biodegradable plastics from activated sludge is a promising sustainable technology. In this study, polyhydroxybutyrate (PHB) – a biodegradable plastics material was produced by activated sludge performing enhanced biological phosphorus removal (EBPR) in batch experiments under anaerobic, aerobic and anaerobic/aerobic conditions. Under anaerobic conditions, the PHB content of 28.8% o...

Volatile fatty acids influence on the structure of microbial communities producing PHAs

Polyhydroxyalkanoates (PHAs) can be produced by microorganisms and are a biodegradable alternative to fossil-fuel based plastics. Currently, the focus is on reducing production costs by exploring alternative substrates for PHAs production, and on producing copolymers which are less brittle than monomers. Accordingly, this study used a substrate consisting of wastewater from waste-glycerol ferme...
